PRESS RELEASE ReNeuron presents positive pre-clinical efficacy data with ReN009 stem cell therapy for peripheral arterial disease at leading US scientific conference Guildford, UK, 16 November 2009: ReNeuron Group plc (LSE: RENE.L) announces further positive pre-clinical efficacy data with its ReN009 stem cell therapy for peripheral arterial disease (PAD). PAD is a chronic and debilitating disease that progressively restricts blood flow in the limbs, causing cramping, chronic pain and in extreme cases, loss of limb. PAD is commonly associated with other conditions such as diabetes, obesity and stroke. At least 1 in 20 people over the age of 55 have some degree of PAD and it becomes more common with increasing age. The research was conducted by Professor Paolo Madeddu, Chair of Experimental Cardiovascular Medicine and colleagues at the Bristol Heart Institute, University of Bristol, UK, and builds on earlier preliminary work conducted by that group. In two studies, the researchers tested ReNeuron's lead CTX neural stem cell line, via intramuscular injection, in a recognised murine hind limb ischaemia model. A range of doses were used across the studies, from 3,000 to 3 million cells per treatment. A significant and dose-dependant recovery of blood flow to the ischaemic limb was seen as soon as three days after ischaemia, with significantly increased revascularisation of the damaged tissue after 21 days, as measured by increased capillary and arteriole density. The results of the above studies were presented by poster at the American Heart Association Scientific Sessions 2009 in Orlando, Florida, running from 14-18 November. An AHA Scientific Council travel stipend has been awarded to the Bristol team poster presenter in recognition of the scientific excellence of the abstract submitted to the Scientific Sessions. ReNeuron is developing its ReN009 therapy as an allogeneic (non-patient specific) stem cell treatment for late-stage PAD, or critical limb ischaemia, in diabetic patients for whom PAD is a side-effect of their diabetes. Further pre-clinical studies are currently in progress with ReN009, focusing on diabetic models and utilising the newly-developed frozen formulation of the CTX cell line which will be used in future clinical trials. The results of these further studies will be reported in early 2010. In the meantime, ReNeuron has scheduled meetings within the next month with both the UK Medicines and Healthcare Regulatory Agency (MHRA) and the UK Gene Therapy Advisory Committee (GTAC) in order to discuss and agree certain aspects of the late pre-clinical development plan for the ReN009 therapy. Equivalent meetings are also planned with regulatory authorities in other territories, including the US, ahead of initial clinical trials expected to commence in 2011. Commenting on the results of the study, Professor Madeddu said: "We are excited by these latest results using the CTX stem cell line, where we have successfully replicated, in a dose-dependent way, the results of a promising initial study in a recognised hind limb ischaemia model. We see real clinical potential in this cell line in ischaemia, where the angiogenic properties of the cell line appear to play a significant role in restoring blood flow in the affected region." ReNeuron announces publication of new research using ReNcell® stem cell products Guildford, UK, 2 October 2009: ReNeuron Group plc (LSE: RENE.L) announces the recent publication of new research using its ReNcell® neural stem cell products. These human cell lines and supporting products are marketed through US-based Millipore Corporation for academic and commercial research. Researchers from the University of California, San Diego School of Medicine and the Salk Institute for Biological Studies, La Jolla, California have developed a safe strategy for reprogramming cells to a pluripotent state without the use of the potentially harmful viral delivery mechanisms or genomic insertions that were necessary in earlier cell reprogramming studies conducted elsewhere. In the study, researchers were able to reprogramme ReNeuron's ReNcell®VM cell line using a single factor to generate stable human induced pluripotent stem cells (iPS cells), similar to human embryonic stem cells with the ability to differentiate into any type of cell in the body. The current excitement surrounding iPS cells lies in their potential to circumvent the need to extract such pluripotent stem cells from the early human embryo, as is the case with human embryonic stem cells. The research paper describes the ReNcell®VM cell line used in the study as representing a reliable, homogenous and commercially available tool to dissect the individual factors required for cell reprogramming, and a potentially better standard model for fundamental cell reprogramming studies than human primary fibroblasts, a cell type typically used in this type of research. Moreover, the paper describes the reprogramming strategy used in the study, due to its lack of viral or transgene interference with the cells, as representing a safe way to generate iPS cells for potential clinical use as well as basic research. The research, led by UCSD Stem Cell Program researcher Alysson Muotri, PhD, assistant professor in the Departments of Pediatrics at UC San Diego and Rady Children's Hospital, San Diego and UCSD's Department of Cellular and Molecular Medicine, was published in the on-line journal, PLoS One in September (Marchetto, M.C.N. et al. (2009) PLoS ONE 4(9), e7076). Dr John Sinden, Chief Scientific Officer of ReNeuron, said: "This latest publication is a clear example of how ReNeuron's ReNcell® lines can be successfully used at the very cutting edge of stem cell research. In this study, the ReNcell®VM cell line was proven to be efficient in generating stable iPS cells at a significantly higher frequency than that obtained using human primary fibroblasts. Furthermore, the study showed for the first time that the iPS cells generated carried a transcriptional "signature" of the original ReNcell® donor cells, raising interesting questions as to what effects, if any, this "memory retention" might have. 2009 AGM trading update Guildford, UK, 17 September 2009: ReNeuron Group plc (LSE: RENE) today announces a business update ahead of its 2009 Annual General Meeting. ReN001 stem cell therapy for stroke In July, the Company announced that the UK Gene Therapy Advisory Committee (GTAC) had issued a final ethical opinion, subject to certain conditions, in favour of ReNeuron's proposed first-in-man clinical trial with ReN001, its stem cell therapy for stroke. GTAC acts as the national research ethics committee for gene therapy and stem cell therapy clinical trials in the UK. This final ethical opinion followed regulatory approval for the clinical trial granted by the UK Medicines and Healthcare Products Regulatory Agency (MHRA) earlier in the year. The ReN001 Phase I trial will take place in Scotland at Glasgow's Southern General Hospital, in patients who have been left disabled by a stroke. Since the July announcement, the Company has been continuing its interactions with both GTAC and the MHRA to clarify and address the conditions attaching to the favourable ethical opinion. These conditions took the form of amendments to the original clinical trial protocol and the provision of supplemental, non-safety-related pre-clinical data. The Company is pleased to announce that it has now received regulatory approval from the MHRA for the proposed clinical trial protocol amendments. The Company is now working with GTAC to address the remaining points by way of an expedited "real-time" review process outside of the formal GTAC meeting schedule. The Company aims to meet all remaining conditions and complete all remaining pre-trial preparation activities in time to allow commencement of the ReN001 clinical trial in late 2009 or early 2010. Other developments The Company continues to make good progress with its ReN009 stem cell therapy for peripheral artery disease (PAD), in collaboration with the UK's Bristol Heart Institute. PAD is a chronic and debilitating disease that progressively restricts blood flow in the limbs, causing cramping, chronic pain and in extreme cases, loss of limb. The disease is commonly associated with other conditions such as diabetes, obesity and stroke. Following the presentation of initial positive pre-clinical efficacy data from this collaboration earlier in the year, the Company intends to announce further data from its research collaboration with the Bristol team shortly. The Company is engaging a number of pre-clinical and clinical practitioners in the PAD field in the UK, Europe and the US to advise and assist the Company in guiding the ReN009 therapy through to the clinic. The Company will also commence dialogue with the regulatory authorities in these territories shortly, in order to clarify the optimal pathway to the clinic for the ReN009 therapy. Due to the nature of the disease, the type of therapy and the pre-clinical data package that already exists for the CTX stem cell line being used in this indication, the Company expects the remaining pre-clinical and subsequent clinical pathways for ReN009 to be considerably shorter, less challenging and less costly than those for the ReN001 stroke therapy. Subject to further regulatory advice, the Company is targeting the commencement of clinical trials with ReN009 in 2011. A key factor driving progress with ReN009 has been the Company's recent breakthroughs in the reformulation and manufacturing process for the CTX stem cell line. ReNeuron recently announced a technology transfer and manufacturing contract with Angel Biotechnology plc to generate clinical-grade lots of a second-generation formulation of the CTX cells for subsequent pre-clinical and clinical use in the Company's ReN001 and ReN009 programmes. The practicalities associated with distributing whole cell treatments are a key consideration in the commercialisation of cell-based therapies such as ReNeuron's. This new freeze-thaw formulation makes possible the scale-up and production of CTX cells for wide-scale use across the globe, both clinically and commercially. This scale-up capability should therefore provide the Company with a key advantage when seeking commercial development partners for its therapies. Sales of ReNeuron's ReNcell® range of cell lines and reagents for non-therapeutic applications continue to increase via the Company's out-licensing agreement with Millipore Corporation. Millipore recently secured its first commercial research sub-license for the ReNcell® products with a leading European pharmaceuticals group. Other major companies and academic researchers continue to show interest in the ReNcell® products, with ReNeuron deriving royalty income from each sale made by Millipore. Financing The Company continues to keep tight control over its costs, with current cash resources projected to last well into the second quarter of 2010. The Company will look to seek further equity-based funding at the appropriate time and is also actively pursuing a number of non-dilutive governmental, regional and charitable translational funding sources, such as the UK Technology Strategy Board's recently announced programme to fund commercial research and development projects in regenerative medicine. RNS Number : 8087Y ReNeuron Group plc 10 September 2009 ReNeuron signs manufacturing contract for second-generation CTX stem cell product Guildford, UK, 10 September 2009: ReNeuron Group plc (LSE: RENE.L) today announces that it has signed a contract with Angel Biotechnology Holdings plc ("Angel"), under which Angel will manufacture clinical-grade lots of a second-generation formulation of ReNeuron's lead CTX stem cell line, for subsequent pre-clinical and clinical applications. The new formulation enables the CTX cells to be readily frozen down for shipping and storage, and thawed at the point of use. This process enables a greatly enhanced shelf life for the product and the capability to ship the cells for clinical and commercial application anywhere on the globe. In due course, the Company plans to utilise this enhanced CTX cell formulation for multi-centre, proof-of-concept clinical trials with its ReN001 stem cell therapy for disabled stroke patients, as well as for late pre-clinical testing and initial clinical trials with its ReN009 stem cell therapy for peripheral artery disease.
